    Neural Substrates of Reliability-Weighted Visual-Tactile Multisensory Integration

    As sensory systems deteriorate in aging or disease, the brain must relearn the appropriate weights to assign each modality during multisensory integration. Using blood-oxygen level dependent functional magnetic resonance imaging of human subjects, we tested a model for the neural mechanisms of sensory weighting, termed “weighted connections.” This model holds that the connection weights between early and late areas vary depending on the reliability of the modality, independent of the level of early sensory cortex activity. When subjects detected viewed and felt touches to the hand, a network of brain areas was active, including visual areas in lateral occipital cortex, somatosensory areas in inferior parietal lobe, and multisensory areas in the intraparietal sulcus (IPS). In agreement with the weighted connection model, the connection weight measured with structural equation modeling between somatosensory cortex and IPS increased for somatosensory-reliable stimuli, and the connection weight between visual cortex and IPS increased for visual-reliable stimuli. This double dissociation of connection strengths was similar to the pattern of behavioral responses during incongruent multisensory stimulation, suggesting that weighted connections may be a neural mechanism for behavioral reliability weighting

    Exogenous coenzyme Q10 modulates MMP-2 activity in MCF-7 cell line as a breast cancer cellular model

    <p>Abstract</p> <p>Background/Aims</p> <p>Matrix Metalloproteinases 2 is a key molecule in cellular invasion and metastasis. Mitochondrial ROS has been established as a mediator of MMP activity. Coenzyme Q<sub>10 </sub>contributes to intracellular ROS regulation. Coenzyme Q<sub>10 </sub>beneficial effects on cancer are still in controversy but there are indications of Coenzyme Q<sub>10 </sub>complementing effect on tamoxifen receiving breast cancer patients.</p> <p>Methods</p> <p>In this study we aimed to investigate the correlation of the effects of co-incubation of coenzyme Q10 and N-acetyl-L-cysteine (NAC) on intracellular H2O2 content and Matrix Metalloproteinase 2 (MMP-2) activity in MCF-7 cell line.</p> <p>Results and Discussion</p> <p>Our experiment was designed to assess the effect in a time and dose related manner. Gelatin zymography and Flowcytometric measurement of H2O2 by 2'7',-dichlorofluorescin-diacetate probe were employed. The results showed that both coenzyme Q10 and N-acetyl-L-cysteine reduce MMP-2 activity along with the pro-oxidant capacity of the MCF-7 cell in a dose proportionate manner.</p> <p>Conclusions</p> <p>Collectively, the present study highlights the significance of Coenzyme Q<sub>10 </sub>effect on the cell invasion/metastasis effecter molecules.</p

    Skin and soft tissue infection in people living with human immunodeficiency virus in a large, urban, public healthcare system in houston, Texas, 2009-2014

    Antecedentes. Las infecciones de la piel y los tejidos blandos (SSTI) afectan de manera desproporcionada a los pacientes con el virus de la inmunodeficiencia humana (VIH). Se han observado disminuciones recientes en la incidencia de SSTI en la población sin VIH. Buscamos estudiar la epidemiología y microbiología de las SSTI en una población de 8597 pacientes seguidos para la atención primaria del VIH en un gran sistema de condado urbano desde enero de 2009 hasta diciembre de 2014. Métodos. Las SSTI se identificaron a partir de la historia clínica electrónica mediante el uso de códigos de facturación de la Clasificación Internacional de Enfermedades-9. Se revisaron los gráficos para confirmar el diagnóstico de SSTI aguda de cada paciente y los datos de cultivo y susceptibilidad abstractos. Calculamos las incidencias anuales de SSTI mediante regresión de Poisson con agrupamiento por paciente. Resultados. Se identificaron 2202 SSTI. De 503 (22,8%) SSTI cultivados, 332 (66,0%) recuperaron Staphylococcus aureus como patógeno, de los cuales 287/332 (86,4%) presentaron S. aureus como único organismo aislado. Entre los aislamientos de S. aureus que mostraron susceptibilidad a los antibióticos, 231/331 (69,8%) fueron resistentes a la meticilina y la proporción no varió por año. La incidencia observada de SSTI fue de 78,0 por 1000 personas-año (intervalo de confianza del 95% 72,9-83,4) y disminuyó de 96,0 infecciones por 1000 personas-año en 2009 a 56,5 infecciones por 1000 personas-año en 2014 ( P<0,001). Otros predictores significativos de la incidencia de SSTI tanto en análisis univariados como multivariados incluyeron un recuento bajo de CD4, una carga viral alta y no ser hispano de habla hispana. Conclusiones. Las SSTI siguen siendo un problema importante en los pacientes ambulatorios que viven con el VIH, aunque las tasas de SSTI parecen haber disminuido en aproximadamente un 40% entre 2009 y 2014.Background. We sought to study the epidemiology and microbiology of SSTIs in a population of 8597 patients followed for HIV primary care in a large, urban county system from January 2009 to December 2014. Methods. SSTIs were identified from the electronic medical record by use of International Classification of Diseases-9 billing codes. Charts were reviewed to confirm each patient’s diagnosis of acute SSTI and abstract culture and susceptibility data. We calculated the yearly SSTI incidences using Poisson regression with clustering by patient. Results. There were 2202 SSTIs identified. Of 503 (22.8%) cultured SSTIs, 332 (66.0%) recovered Staphylococcus aureus as a pathogen, of which 287/332 (86.4%) featured S. aureus as the sole isolated organism. Among the S. aureus isolates that exhibited antibiotic susceptibilities, 231/331 (69.8%) were methicillin resistant, and the proportion did not change by year.     Dynamic immune markers predict HIV acquisition and augment associations with sociobehavioral factors for HIV exposure

    Summary: Prior studies attempting to link biomarkers of immune activation with risk of acquiring HIV have relied on cross sectional samples, most without proximity to HIV acquisition. We created a nested case-control study within the Sabes study in Peru, and assessed a panel of plasma immune biomarkers at enrollment and longitudinally, including within a month of diagnosis of primary HIV or matched timepoint in controls. We used machine learning to select biomarkers and sociobehavioral covariates predictive of HIV acquisition. Most biomarkers were indistinguishable between cases and controls one month before HIV diagnosis. However, levels differed between cases and controls at study entry, months to years earlier. Dynamic changes in IL-2, IL-7, IL-10, IP-10 and IL-12, rather than absolute levels, jointly predicted HIV risk when added to traditional risk factors, and there was modest effect modification of biomarkers on association between sociobehavioral risk factors and HIV acquisition

    Effect of Aqueous Extract of Elaeagnus angustifolia Fruit on Experimental Cutaneous Wound Healing in Rats

    The present study was conducted to investigate the histological changes and wound healing effect of aqueous extract of Elaeagnus angustifolia. After creating full-thickness skin wounds on the back of 45 male Sprague-Dawley rats they were randomly divided into three groups. Treated group received the extract, positive control group were treated with mupirocin ointment 2% and control group did not receive any treatment. Wound healing rates were calculated on days 3, 5, 8, 10, 12 and 15 post-wounding and the wound tissues were harvested at 5, 10, and 15 days for histological analysis and hydroxyproline content measurement. The results indicated a significant increase in the percentage of wound contraction and hydroxyproline content in the treated group comparing to the control and positive control groups. A significant increase in the assigned histological scores was observed at 10 and 15 days in the treated and positive control groups compared to the control group. The results demonstrate that aqueous extract of Elaeagnus angustifolia accelerates cutaneous wound healing, and its effect may be due to the increased re-epithelialization and collagen deposition in wound and so it can be considered as a therapeutic agent for wound healing
